Human Embryonic Kidney Cells

In figure A: Human embryonic kidney cells were transfected with constructs for carrying out this experiment. Immuno-precipitation was carried out of tagged PAG with an antibody against regulator Myc, its interaction is studied here with SRC & BRK follwed by immunoblotting analyzation. The result of this experiment was delaying in electrophorectic mobility of tagged PAG protein, when it was co-expressed with SRC. This delay is considered due to hyper-phosphorylation. However, on co-expression of PAG and BRK this band shift was less evident. In figure C: Co-transfection of HEK 293 cells with MYC-PAG and SRC followed by treatment in the absence or presence of SRC kinase inhibitor SU6656 (5 M) for atleast 1 h. PAG was immunoprecipitated by antibody against MYC, and the binding of CSK was compared by immunoblotting. The disturbance of link b/w CSK and PAG was observed on weakening SRC activity by a small molecular inhibitor SU6656, this experiment illustrated the importance of SRC kinase activity for functioning of CSK. B) to test the same hypothesis with controls, take breast cancer frozen samples of different patients, divide the sample in two groups one will be the test group other the control group. The control group will be given doses of anti tumor medication (say tamoxifen for breast cancer). RNA will be extracted from both groups using trizol and will be followed by purification assay. The breast cancer cell lines will be transfected with empty vectors or pcDNA3-MKP3-V5. Further the transfectants will be placed in media MEM along with phenol red. Followed by SDS PAGE electrophoresis, and transfer to nitrocellulose membrane. After transferring they will be incubated with primary antibody for an hour or above then with secondary antibody to observe the chemiluminescence with a reagent. The cells will then be lysed in a buffer, phosphatase reaction carried out will be observed via assays and transfected cells will be compared with control ones to whom tamoxifen was given.

The questioning of the librarian brings a scolding that is whispered, but full of indignation (Brophy, 2). In Psycho, there are similar aspects of the film that brings this film to life. The most obvious, of course, was Norman Bates and his â€Å"mother.† Bates has a pitch-perfect imitation of his mother, so much so that the audience could never suspect that the mother actually wa s not a separate person at that point. The arguments that emanate from the spooky old house on the hill that is directly behind the Bates Motel could never, in the audience’s wild imagination, be just one person arguing essentially with himself. The voice inflections and the pitch of the voice of the mother are so real that it is that much more shocking to find out, in the climax, that Norman’s mother had been dead for many years, and that the â€Å"mother† was, at that point, nothing but a skeleton with a robe and wig on, sitting in the apple cellar. This is not the only vocal performance that brings suspense to the film. Janet Leigh’s performance as the lead character, Marion, also brings a sense of suspense to the film. For instance, when she is stopped by the policeman early on in the film, her talking is way too fast. This, combined with her overall shifty-eyed look gave the policeman reason to suspect that she was hiding something, and she was, inde ed - $40,000 that she had embezzled from her employer. This same tone of voice is employed when she went to buy a new car – her speech was tight and quick. She was extremely nervous, and this nervousness was evident in her voice and her overall body language. Because the audience knew that Marion was not that good of an actress (although Leigh certainly was), it became obvious when she felt comfortable and when she felt uncomfortable. It was completely in her voice. Therefore, when she was talking to Norman in his room – while she ate her sandwich, while looking at the creepy stuffed owls and such – it was obvious that she felt completely comfortable. For whatever reason, there was nothing in Norman’s weird demeanor that gave her pause. If there was something, Marion would not have been able to hide it. She couldn’t hide nervousness, this was obvious. Therefore, the audience was clued into Marion’s state of mind throughout the film – it showed in her voice and her body language. Similarly, Brophy stated that this is an important part of the vocal characterizations – the changes in the voice of the character would signify the different states of mind that the character might go through during the course of the film (Brophy, 3). The new law governing the formation of cartels ensures that such cartels do no harm to the society and the economy. Histomorphometeric Study of Lymphoid Nodules of Vermiform ABSTRACT Objective: To study the lymphoid tissues of vermiform Appendix at various age groups to establish that appendix is an important organ of gut associated lymphoid tissue (GALT) and its role in etiology of acute appendicitis. Study Design: Comparative study Place and duration: The study was conducted at Islamic International Medical College Rawalpindi from July 2011 to June 2013. Materials and Methods: Four age groups spacing 15 years between each group, except last group (advanced age) which had no limit because of less availability of the specimens, were made. In each group histological sections of 10 normal looking surgically amputated vermiform appendices were included. The parameters including number of lymphoid nodules and diameter of lymphoid nodules measured under the microscope. Results: There was gradual decrease of mean number of lymphoid nodules with advancing age but even at the age of 74 few numbers of lymphoid nodules were observed. There was great variation of diameter of lymphoid nodule at different age groups but at advanced age it was much decreased. Conclusion: The lymphoid nodules decreased with advancing age but these were present even at the age of 74 years. The diameters of lymphoid nodules are remarkable except at advanced age. Introduction The clinical presentations associated with this blind ending tube (vermiform appendix) are so many and diverse that many lives have been lost in the past either due to misdiagnosis, mismanagement or complications associated with it. The appendix length varies from 2 to 20 cm with an average of 9 cm.1 The variable number of lymphoid nodules located in the lamina propria extending to sub-mucosa, are arranged circumferentially around the lumen of the appendix. The crypts of Lieberkuhn are absent between epithelial lining and lymphoid nodules at places where the lymphoid nodules are close to epithelial lining. Zahid A. observed that the vermiform appendix is an important component of mammalian mucosal immune function, particularly B-lymphocyte-mediated immune responses and T-lymphocytes derived extra-thymically and it is also suggested that vermiform appendix is not a vestigial organ.2 The normal function of appendix probably helps to promote local immunity.3 The appendix contains both isolated and aggregated lymphoid follicles and it is a part of gut associated lymphoid tissue (GALT).4 The irregularity of the lumen of vermiform appendix is due to mucosal folds which appear due to lymphoid follicles.5 The lymphoid nodules appear in the vermiform appendix about two weeks after birth6. In full term human fetus there is no obvious lymphoid tissue seen in the vermiform appendix but two well organized lymphoid nodules are observed within first two weeks and about 10-12 well organized and prominent lymphoid nodules are visible in transverse section of vermiform appendix of man of thirty two weeks old.7 As the age advances, the weight of the appendix decreases because number and size of lymphoid nodules reduces. In or near appendix, in any pathology, the primary and secondary immune response is high in young age. Materials and Methods This study was conducted at Islamic International Medical College Rawalpindi from July 2011 to June 2013. In this study, a total of 40 normally looking surgically amputated vermiform appendix specimens were included. According to age, these specimens were divided into four equal groups. Each group consists of 10 specimens with spacing of 15 years between each group except the last group. Due to less availability of appendices, the last group ranges from 46-74 years. The groups were as follow: Group A: 0- 15 years Group B: 16-30 years Group C: 31-45 years Group D: 46-74 years The 5 micrometer thickness sectioning of tissue blocks with microtome made. The specimens stained with Haemotoxylin and Eosin. Only the results of middle part of the vermiform appendix are included in this study. The histological slides were examined under the light microscope after calibration of ocular micrometer. Procedure for measurement of the diameter of the lymphoid follicle The measurements were taken twice for each nodules, one was taken among the maximum transverse diameter and another at perpendicular to the first one. Then the number of micrometer divisions were multiplied by the correlation factor derived earlier keeping the magnification constant. Diameter of the lymphoid follicle was calculated as: Maximum transverse Diameter + Perpendicular diameter Diameter of lymphoid follicle = 2 By using the above formula the diameter of all lymphoid nodules of each slide were measured and then mean diameter of each slide calculated. Then the mean diameter of each group calculated as shown in the tables. Discussion At different age groups, the microanatomy of appendix is variable. The lumen size and wall thickness depends on number and diameter of lymphoid nodules. There are large numbers of lymphoid nodules in the vermiform appendix and it is a part of Gut Associated Lymphoid Tissue (GALT). It is proposed by Zahid A. that appendix perform to train immune system of the body for production of antibodies when exposed to antigen and also play a role in foetal development by manufacturing hormones.2 In this study, the highest number of lymphoid nodules (10.3) is in the group B (16- 30 years) and the lowest number of lymphoid nodules (3.8) is in the group D (46-74 years). In a Bangladesh study8, the highest number of lymphoid follicles was in age group up to 20 years and lowest in age group between 56-70 years and similarly the diameter of lymphoid follicles is highest in age group up to 20 years and lowest in age group of 56-70 years but the age grouping was somewhat different from our study. In another study, the highest average diameter of lymphoid follicles was found in group 0-20 years and lowest average diameter was found above 50 years age.9 The above referred study agreed with Borley5 and Arey.10 In another Bangladesh study11 the mean numbers of lymphoid nodules are less than half of our study. In old age, the numbers of the lymphoid nodules are reduced and these are replaced by fibrous tissue.11,12 It is observed in our study, that although the mean number of lymphoid nodules is lowest (3.8) in group D (45-74Years) but even at the ages of 60 years, 63 years, and 65 years, the number of lymphoid nodules are equivalent to some specimens of group A, B, and C. Even at the age of 74 years three lymphoid nodules were found. Although the lymphoid nodules and other structures in the wall of appendix are replaced by fibrous tissue in old age but normally it is also observed in many adults13. The germinal centers in most of the lymphoid nodules of human appendix are present in the children up to16 years of age but the number of germinal centers are less in adults and they are absent in the nodules in old age persons.14 In our study the germinal centers were not explored. Like the vermiform appendix, the Peyer’s patches (Lymphoid nodules) which are concentrated at distal 25cm of ileum are more numerous in young adults and they become less prominent as age advances.15, 16 Conclusion The lymphoid nodules decreased with advancing age but these were present even at the age of 74 years. Timothy Leary "Turn on, tune in, drop out." That saying has turned into the slogan of Timothy Leary’s mind-expanding movement. Although a graduate of both West-Point and Berkley, and a Harvard professor, these were not his greatest lifetime achievements. Throughout his publicized life, he became the spokesperson of the psychedelic age. His devotion to the belief that LSD and marijuana were gateways to enlightenment resulted in a new church, numerous prison sentences, and a following of both celebrities and the general public. When people think of Timothy Leary their immediate response is "Turn on, tune in, drop out," his trademark line, although the meaning of it has often been misinterpreted. Playboy Magazine had thought that his message was advocating, "getting high and dropping out of school," (Marwick 311). When asked by the magazine to explain the meaning of the phrase he responded, " ‘Turn on’ me ans to contact the ancient energies and wisdoms that are built into your nervous system. They provide unspeakable pleasure and revelation. ‘Tune in’ means to harness and communicate these new perspectives in a harmonious dance with the external world. ‘Drop out’ means to detach yourself from the tribal game." (Marwick 312). This was not the first time his methods were questioned. Leary was first publicly noticed, and criticized by then fellow Harvard professors, for his interest in LSD when he and friend, Robert Alpert, wrote an article for the Bulletin of Atomic Scientist. In the article they described a circumstance that in the event of war, the Russians might try to lace the American water supply with LSD. Then, when everybody in America is stoned, the Russians would seize power. They explained that in order to prevent the scenario from happening, everyone should take a dose of LSD so they can get used to the effects (Sterns 278). Although the a rticle shocked the Harvard staff, it didn’t cause him to get fired. Leary was dismissed from Harvard in 1962, only four years after he began teaching there. Leary had experimented with psilocybin, a mind-altering chemical, on his own and the university repeatedly asked him to stop, he refused (Brash 139). Although it angered university 2. authorities, they couldn’t do anything more about the subject since it was a legal substance. His last words were "Why not?" and after his death his long time friend, John Barlow, wrote, "Timothy Leary died unashamed and having, as usual, a great time. He made good on his promise to ‘give death a better name or die trying.’ " (Marwick 345). Timothy Leary has influenced many people throughout his life by his contribution to society. Whether he advocated the use of drugs or not doesn’t make a difference, although the are what stuck out in people’s minds. The greater message that his life taught was the will of man, and the lengths a person will go to in order to get the point across to the masses. His focus was not money, he gave that up when he lost his job at Harvard, but in his belief that he was right. He truly believed that LSD could enlighten people, and his intentions were to help. A powerful debate whether the middle class is essentially defined by cultural or economic factors still remains an issue. A rich tradition is devoted to disentangling economic from cultural components of a class. According to Karl Marx, the middle class is an outgrowth of economic factors, primarily capitalism. Many people tend to disagree with Marx that capitalism is the only important factor in the outgrowth of the middle class. Judith R. Blau argues that her understanding of the middle class has much to do with inclusive cultural values. Blau demonstrates her opinion though her ethnography, Social Contract and Economic Markets. I believe that Karl Marx’s economic factors and Judith Blau’s cultural factors together d efine the middle class. Karl Marx believed class was a matter of economics, that is, how the individual fits into the pattern of modern capitalist society. Marx argued that the whole of capitalist society was constructed in order to support this idea including the society’s infrastructure. Marx believed that social classes arise when a group gains control of the means of production. This group also has the power to maintain or increase its wealth by taking advantage of the surplus value of labor. Many people question why a worker would labor under such conditions. The reason is quite simple according to Marx. The reason is political and social representation. Members of this class elect representatives who pass laws that serve their interests. Landlords and factory owners were able to use their control of resources to exploit the unlanded laborers in the newly emerging factories. Karl Marx looks at human societies as a whole, and asks how they reproduce themselves, and as a result, change. For Marx a fundamental question about any society is whether it can produce more than it needs to reproduce itself, that is, a surplus product. Karl Marx believed that the middle class is based upon economic factors and rooted in solely that perspective. Without these referrals and repeat business these companies would have to rely a lot on marketing and advertising strategies. My reasoning behind my choices for these businesses that would not benefit from the five questions is because these companies are very well expanded. With many stores in many different locations of one state and through-out many states these companies do not rely on word of mouth or repeat business. As their marketing strategies, advertising strategies, and sales are what continuously brings in repeat customer, and new customers.

The institutions that supported early capitalism were banks and joint-stock companies such as the English East India Company and their Dutch counterpart. 5. The Dutch and English supported the growth of capitalism because the English East India Company and their Dutch counterpart were two large trading companies that spread the ideas of capitalism on a larger scale than ever before. . The Joint-stock companies set p by the ideas of capitalism and the English/Dutch would be authorized to conquer, explore, and colonize distant lands in search of commercial opportunities. Therefore, early capitalism developed in the context of Imperialism, as European peoples established fortified trading posts in Asia and colonial regimes in both South east Asia and the Americas. Imperial expansion was important to the spread of capitalism, since it enabled European merchants to get natural resources that would be distributed. 7.Crafts and guilds had fixed prices and wages and they regulated standards of quality. They did not seek to realize profits, therefore discouraging competition and resisted technological innovation. 8. The putting out system was system in which capitalist entrepreneurs delivered unfinished materials to rural households where the artisans would finish a part of the product in their homes (similar to an assembly line) and pass it on until the product was finished and then it was giving to the entrepreneurs, and then the artisans were paid for their labor. 9.The advantages of the putting out system were that because of the plethora of rural labor, entrepreneurs spent relatively title on wages and profited greatly. 10. The putting out system introduced large sums of money to the countryside which brought materi al benefits and disrupted the long-established patterns of rural life. The standard of rural life rose dramatically; households acquired more goods and rural people wore nicer clothes, ate better food, and drank better wine. Individuals had the opportunity to become wealth and stop relying on neighbors and family. Young adults and women began to earn their own incomes. 11.In Eastern Europe lacked cities so there was no alternative to working in the countryside, o landlords took advantage on this situation by forcing peasants to work under extremely harsh conditions whereas in Western Europe there were better working conditions and more cities. Western Europe was far better in comparison to Eastern Europe in the area of labor. 12. Capitalism posed moral challenges. Medieval theologians believed that profit-making was morally dangerous, since profiteers looked to their own advantage rather than the welfare of the larger community. The Church considered interest an unearned and immoral profit. 13.Adam Smith's response was that society loud prosper when individuals pursued their own economic interests. 14. Although Capitalism may not have necessarily caused changes in family life, it encouraged developments that helped to define the nature and role of the family. Capitalism favored the nuclear family; it offered opportunities doe independent families to increase their wealth. As these families became more important economically, their importance socially and emotionally increased. Love became more important in marriage (not simply strengthening alliances) and parents and their children became more important to family life.

Finishing school is the beginning of the independent life for millions of school graduates. Many roads are open before them: technical schools, colleges, universities. But it’s not an easy thing to choose a profession out of more than 2,000 existing in the world. That’s why the topic of choosing the career path becomes one of the major questions for many students. Some people know ride away what they want to do in life, for others it’s an extremely difficult decision.Selecting the right career path, you need to be proactive about your decision because this choice will change your entire life! Sometimes it’s difficult to establish career goals if you’re not really sure what you’d like to do. A good place to start is some self examination. First of all you should decide what are your or could be your talents. These are the things that you are naturally good at. They can be discovered and developed, but not taught. Secondly, concentrate on your skills – these are how you do something. They can be learnt and are transferable.The extent to which you can develop your skills can be influenced by your talents. Finally, determine your character traits – whether you like dealing with people, what mental stimulation you need, whether you like physical work, your emotional make-up, how you handle stress and other emotions, what sort of environment you would like to work in. You might also want to look at what sorts of roles, industries, or areas interest you. Make sure you’re honest, keep an open mind so you consider all options before you make your decision.You also should be careful not to pursue ideas or careers that seem attractive in some way (well paid), but that aren’t actually suitable for you or in which you don’t have the natural strengths or personality type. Everyone of us has a unique set of skills and talents, so you should focus on them, and then think about where and how to apply t hem. To narrow your search down, you may like to create a list of the careers that you like and you are suitable for. This way, there is the lower chance of having hard times in your life – you can totally rely on them. In this situation, I believe, is it fair to choose a job that gives you personal satisfaction even if the salary is lower than other available jobs. However, if you don’t have a family support, you shouldn’t concentrate just on making money.It’s very important to enjoy yourself. The job you do, makes a big difference in your everyday life. Who wants to be nervous and irritated every s ingle day after a boring day at work? This can be caused by the wrong decision in choosing the career path. That’s why, on one hand you should be sensitive to your family members’ suggestions, but on the other – â€Å"Listen to your dreams – those are the sounds no one else can hear. † (Kobi Yamada) Another key to happiness and satisfaction in your life is balance. Finding it can be a bit challenge to a lot of people.But if you know how to balance your professional and personal lives, you will have more time to do the things that you want to do! First of all, you should prioritize (decide what things are the most important to you). Do your best to improve your personal skills. Secondly, manage your time and divide your day into smaller parts to create some sort of schedule. This will also put more limits and make you work more productively. In addition to all of this, you should enjoy yourself. If you do, then you will not feel stressed and burned out.This way, you can get out of your office happy and ready to face your personal life. The balance in your professional and personal lives is a mental state that is reached, when you do not have the need to separate these two worlds. The Who

The Who is considered one of the most powerful rock bands of the 21st century. They started their band in London, England. The Who formed in 1964. They sold over 100 million records worldwide. They were inducted into the Rock and Roll Hall of Fame in 1990. The Who can also be known as The Detours or The High Numbers.Band MembersThe Who developed from an earlier group called The Detours. From there they established their band as a part of the pop art and mod movements. The founding members of The Who were Roger Daltrey, Pete Townshend, John Entwistle and Keith Moon. Daltrey was the lead singer, Townshend was a guitarist and singer, Entwistle was a bass guitarist, and Moon was a drummer. All of the band members grew up around London. Daltrey had a hard time fitting in at school and discovered rock and roll. He founded the Detours in 1959. Townshend's parents were very supportive of him since his dad played the saxophone and his mother had sung the entertainment division during World War II. Just like Townshend, Entwistle also liked rock and roll. He started to play the guitar but since he had large fingers, it was hard for him to play it.Early careerAfter the Detours became the Who, many things changed. Sandom left the band since he wasn't getting along with Townshend. After that, they didn't talk for many years. During a gig at Oldfield, the Who met Keith Moon. Townshend accidentally broke his guitar on stage and he was angered by the audiences laugh. This made him crush the guitar on stage but he picked up another one and continued to play. The next week when the band played again, the audience wanted to see a repeat of what Townshend did. This is how auto-destructive art became part of the Who's live shows.SuccessesThe Who had many successes. Their first single, â€Å"I Can't Explain† reached the UK top ten. Their next single called â€Å"My Generation† was released in October. Although it was originally supposed to be written as a slow blues Entwistle played a bass solo in it. In the end, it turned out to be a very powerful song. The Who's most successful solo release was a Daltrey solo album. Daltrey was a very good singer, but he also found success in acting. The Who also became the first rock and roll act to play Metropolitan Opera House in New York City. Their fourth album, 1969's rock opera Tommy, was a commercial success.Rock and Roll Hall of Fame and AchievementsThe Who was inducted into the hall of fame in 1990. They even have a featured collection in the Rock and Roll Hall of Fame museum which includes a drumhead from 1968. The Who also made it into the UK Music Hall of Fame. They had seven of their albums appear on Rolling Stone's 500 greatest albums which were in 2003. That made the Who have more than any other rock and roll band except four. The Beatles, Bob Dylan, the Rolling Stones, and Bruce Springsteen. The Who even held a world record for being the world's loudest rock and roll bands!End of Career and DeathOut of the four founding members of the Who, Moon and Entwistle both died. Moon died in 1978 because of a drug overdose. Townshend, Daltrey, and Entwistle released â€Å"Who Are You† shortly after his death. The song was based on Moon's death. Sadly, Entwistle died 24 years later in 2002. He had a heart attack at Hard Rock Hotel due to cocaine. This made plans for a new album delayed.ConclusionThe Who has changed many lives with their energetic performances. They have influenced many people. They have even influenced the pop-punk band Panic! At the Disco. The Who have continued to play as a band even after 54 years of being together. They have won many awards for their great recordings. They are not composed by instrument, but they are pretty familiar to us. These sound, after editing, provided us a strong sense of rhythm. Combining the dance, it allows us to think of the deep meaning of each theme. Amount of eight pieces of dance, the one I like the most is Bad Faith. The two performers displayed a strong depressed feeling through their repetitive actions with some change in each repeat. They had a dialogue, but they didn’t act like talking. They rely on each other closely but sometimes they have problems and get separate. They used their bodies to express feeling each time they talked. Such movement is oppressing to me. I love it although it is not as large-scale as others performance I saw before. And because of this reason, I sat really close to the stage and the dancers. I could hear musicians tuning their instruments, I could see light refected on performers’ faces and, the most important thing, I could see dancers’ facial expression. Sometimes they even had no background music. Instead, they used the sound of dancer’s feet rubbing the floor or footsteps as the beat. Attending a live performance is really more enjoyable than watching the same event on television. Dancer is not just a job that repeats movement that other instructs he or she to do, but an artist that use their body as a tool to express emotion, story, or ideas in a nonverbal way. From viewing this concert, I discovered the motion of dance , I feel the change of my mood with the fluctuation of motion performed in different parts of the concert. From these eight pieces of dance, I feel like I have eight times of experience. From this dance, I discover that the dance has its own motion. Jack recognizes the weakness in the group of boys, using their gnawing fear of the beast to turn them to his side, against Ralph. Though much more direct, Jack uses his power to threaten the boys on Ralphs side, such as Samneric, to hail to savagery and chaos, much like Abigail did to Mary. He dominates the island, getting what he wants, and eliminating those such as Piggy and Ralph, who stand in his way. Abigails tactic of lying, manipulating fear and abusing her power in court grants her the same reward of getting her way, and pushing aside enemies like Elisabeth. The only difference is that Abigails actions come with far bigger consequences, more than Parris, Putnam or Danforth, fleshed out on a larger scale of victims who fell in the face of her machinations. In the end, Arthur Millers Crucible is a fine study of manipulation and abuse of power, shown in various forms, through vicious antagonists, always exploiting Salems fear to achieve their own selfish goals and further themselves on the social food chain. What Miller is perhaps attempting to demonstrate through this play is that those in positions of power will always abuse it, especially when faith is involved, because of the manipulation of that faith to create fear and control (Bardem), as have done Danforth, Parris, Putnam and Abigail.
